5-hydroxytryptamine type-3A receptor in the process of fear extinction

چکیده
The 5-hydroxytryptamine type-3 (5-HT3) receptor is the only ligand-gated ion channel in the 5-HT receptor subfamilies. Although it is known that the 5-HT3 receptor is expressed in the hippocampus, amygdala and prefrontal cortex, it is not well studied whether it has roles in fear memory processes. We performed behavioral analyses of 5HT3A receptor knockout mice in fear conditioning tests and found that the 5-HT3 receptor is indispensable for the fear extinction, but not for the acquisition or retention of fear memory. Furthermore, we found the 5-HT3A receptor contributes to the specificity of context in fear extinction processes. Our results indicate the 5-HT3A receptor is one of the key molecules for regulation of fear extinction. Moreover, it could be an important treatment target for the fearrelated disorders, such as post-traumatic stress-disorder (PTSD).

A single amino-acid in the TM1 domain is an important determinant of the desensitization kinetics of recombinant human and guinea pig alpha-homomeric 5-hydroxytryptamine type 3 receptors.
Desensitization of ligand-gated ion channels shapes synaptic responses and provides critical neuroprotection at central synapses, yet the molecular mechanisms underlying the desensitization process are poorly understood. Using the whole-cell voltage-clamp technique, we investigated desensitization kinetics of recombinant human and guinea pig alpha-homomeric 5-hydroxytryptamine type 3 (5-HT(3A))...
متن کاملImpaired Extinction of Learned Contextual Fear Memory in Early Growth Response 1 Knockout Mice
Inductive expression of early growth response 1 (Egr-1) in neurons is associated with many forms of neuronal activity. However, only a few Egr-1 target genes are known in the brain. The results of this study demonstrate that Egr-1 knockout (KO) mice display impaired contextual extinction learning and normal fear acquisition relative to wild-type (WT) control animals. Genome-wide microarray expe...
متن کاملEffects of a Flavonoid-Rich Fraction on the Acquisition and Extinction of Fear Memory: Pharmacological and Molecular Approaches
The effects of flavonoids have been correlated with their ability to modulate the glutamatergic, serotoninergic, and GABAergic neurotransmission; the major targets of these substances are N-methyl-D-aspartic acid receptor (NMDARs), serotonin type1A receptor (5-HT1ARs), and the gamma-aminobutyric acid type A receptors (GABAARs). Several studies showed that these receptors are involved in the acq...
متن کاملSubunit-dependent modulation of the 5-hydroxytryptamine type 3 receptor open-close equilibrium by n-alcohols.
5-Hydroxytryptamine (5-HT, serotonin) type 3 (5-HT(3)) receptors belong to the alcohol-sensitive superfamily of Cys-loop ligand-gated ion channels, and they are thought to play an important role in alcoholism. Alcohols with small molecular volumes increase the amplitude of currents evoked by low 5-HT concentrations and shift the 5-HT concentration-response curve for 5-HT(3) receptor activation ...
متن کاملEffects of Systemic Administration of Oxytocin on Contextual Fear Extinction in a Rat Model of Post-Traumatic Stress Disorder
Introduction: One of the hallmark symptoms of posttraumatic stress disorder (PTSD) is the impaired extinction of traumatic memory. Single prolonged stress (SPS) has been suggested as an animal model of PTSD, since SPS rats exhibited the impaired fear extinction. Oxytocin (OXT) has been recently suggested as a potential pharmacotherapy for treatment of PTSD. In this study, using SPS rats we inve...
